At approximately the same time, across town, the son's wife, a former Broadway musical star, dies from poison.

The curious factor is that the medical examiner cannot identify the way in which the poison was administered to the wife, except to say that no traces were found in the stomach and no marks of a hypodermic are found. Vance attends the son's home, and investigates the wife's death—later that evening, the sister of the son and heir is also poisoned.
When he recovers, the son suggests that his mother may have been responsible for the poisoning, but Vance also finds a note that suggests that the wife committed suicide. There are other characters connected with the family upon whom suspicion falls, including the sister's two suitors, one of whom is the family physician and the other the chief croupier at the family casino, and the children's uncle, who manages the casino. Vance must determine the method by which the poison was administered, and, at the same time, follows a trail that leads to one of the character's research into the production of deuterium , or "heavy water", which had just been discovered in Having worked out the murderer's plot and identity, Vance puts himself at the mercy of the murderer, who is holding Vance at gunpoint, in order to hear a confession—then, the murderer is killed in an exciting climax.

Crime novelist and critic Julian Symons wrote: The Casino Murder Case starred Paul Lukas as Philo Vance, and was a fairly faithful reproduction of the principal details of the novel. When a young woman asks him to help The ghosts of three elderly industrialists killed in an airplane crash return to Earth to help reunite a young couple whom they initially brought together. Brown, because he hasn't been able to get any hard evidence against him.
He then goes after Brown's girlfriend who despises him, for information instead. When Philo Vance receives a note that harm will befall Lynn at the casino that night, he takes the threat seriously while the DA dismisses it.

At the casino owned by Uncle Kinkaid, Lynn is indeed poisoned under the watchful eye of Philo. However, he recovers, but the same cannot be said for Lynn's wife Virginia, who is at the family home.
Only a family member could have poisoned Lynn and Virginia and everyone has their dark motives. Philo will follow the clues and find the perpetrator.

Rosalind Russell saves this movie from being just another '30s mystery out of the mystery machine. Paul Lukas, despite his panache is not a good Philo Vance because of his accent. This is more of a romantic "screwball comedy" type of film than a mystery, but taken in that aspect, it's pretty good. The auction scenes and Alison Skipworth are wonderful.
The chemistry between the unflappable Russell and Mr.
Lukas is really neat, although in the novels romance never got in the way of a good murder. Ernest Heath never got a fair shake. In the books he was never quite the boob portrayed on the screen.

At the end of the bitter quarrel, which involves Mrs. Llewellyn's son Lynn and his wife Virginia Louise Henry , Virginia announces that she has decided to leave the house and go to Chicago. Llewellyn's secretary, are introduced to each other, and Doris immediately takes a liking to Vance. Doris immediately recognizes the return address as being that of the Llewellyn's townhouse in Closter, and notices that the letter was typed on her typewriter.
Vance assigns Sergeant Heath Ted Healy to help stake out the casino that night, but their presence does not prevent Lynn from suddenly collapsing at the card table. At the same time, Doris informs Vance that Virginia has died at the Llewellyn house.
